Study: Single-nucleus resolution mapping of the adult C. elegans and its application to elucidate inter- and trans-generational response to alcohol 41749 cells
Single nucleus RNA-seq of the adult C. elegans non-exposed or exposed to ethanol at the P0 generation and collected at the F1 and F3 generations. Age synchronized fog-1(q253) worms were used to prevent the generation of embryos and therefore only generate data from adult cell types. Worms were cultured on standard nematode growth medium (NGM) plates with OP50 E. coli and maintained at 20°C. The generation of worms to be collected for single-nucleus analysis was moved to 25°C at the L1 stage and grown at 25°C until collection at day 2 of adulthood.
