Performed Drop-seq on stromal vascular fraction from 10 human subcutaneous adipose tissue samples. Subjects were recruited from planned elective procedures from the plastic and bariatric surgery groups at Beth Israel Deaconness Medical Center. All subjects were female, healthy, with stable weight at time of surgery. After patient consent, discarded subcutaneous adipose tissue samples were collected at time of surgical procedure and stromal vascular fraction  (SVF) was immediately processed per modified Rod-Bell dissociation protocol. Pelleted  SVF was initially resuspended and erythrocytes were removed either or antibody-mediated magnetic separation. After erythrocyte lysis, single cell RNA-seq libraries were constructed on individual cells from remaning single cell suspension per the Drop-seq protocol (Macosko et McCarroll). After sequencing, alignment and barcode deconvolution, clustering performed using Seurat 3.1 software.
