We characterize the heterogeneity of CD8 T cells in the tonsils with combined analysis of SMART-Seq2 single cell RNA-seq, FACS intensities, and TCR sequencing on single cells. We find four subgroups of CD8 T cells with different effector phenotypes and analyze the enrichment of tetramer specificity in these groups as well as TCR expansion.
