Nucleus suspensions from uninjured and regenerating pectoral fins at 1, 3 and 7 dpa were fixed and frozen using the Evercode Nuclei Fixation Kit v3 from Parse Biosciences following the manufacturer’s protocol. Fixed and frozen nuclei were then processed to generate barcoded libraries aiming for 7,000 nuclei per sample using the Evercode WT v3 Kit as per manufacturer’s instructions. Resulting libraries were sequenced targeting 20,000 reads per nuclei.
